Udeshi has a strong focus in the area of protein ubiquitination and has developed proteomic methods for enrichment and quantification of ubiquitylation sites at a global scale, to detect ubiquitination from cell or tissue samples. The developed techniques have been critical for the identification of ubiquitin ligase substrates and have revealed key transcription factors modulated upon lenalidomide treatment in myeloma, as well as substrates targeted by recurrently mutated ligases in prostate cancer. Udeshi also uses, develops, and applies methods to identify proteins in a spatial context using enzyme-mediated proximity labeling.
