Rosanna Squitti, PhD, is the Head of the Laboratory of Biology, Department of Laboratory Medicine, Research and Development Division, ‘San Giovanni Calibilita’, Fatebenefratelli Hospital, Rome, Isola Tiberina, Italy. Dr. Squitti was born in Rome, Italy. She received her Master's Degree (‘Laurea’) in Biology (25 May 1993) from the University of Rome ‘La Sapienza’. In 1994 she obtained her Board Certification in Biology. In 1997 she received her Ph.D. in Biology from the University of Rome ‘La Sapienza’. She then moved to the University of Liverpool for a 1-year Post-Doc fellowship. In 2002, she became Head of Laboratory of Biology, Fondazione Fatebenefratelli, AFaR Division – now part of the Department of Laboratory Medicine, Ospedale Fatebenefratelli, Rome, Italy. From 2007 to 2012, she was the Head of the Laboratory of Neurobiology, Department of Neurology at “Campus Biomedico” University, Roma, Italy. In 2009 she obtained her Board certification in Molecular Biology from the National Research Council of Italy (CNR). In 2012 she obtained her MIUR, ASN Professor Board certification in Genetics. In 2016 she was hired as a senior researcher in Molecular Markers Laboratory, IRCCS Istituto Centro San Giovanni di Dio- Fatebenefratelli, Brescia, Italy. In January 2017 she became Voluntary Assistant Professor of Neurology (Visiting Professor), University of Miami, Leonard M. Miller School of Medicine, (Florida, US). Dr. Squitti developed the following international patents: 1) Colabufo, N. and Squitti R, METHOD AND KIT FOR DETERMINATION OF FREE COPPER IN SERUM (WO2014/071973A1); 2) Colabufo N, Berardi F, Leopoldo M, Perrone R, and Squitti R, METHOD FOR DETERMINING FREE COPPER, (WO2014/181215A2). Dr. Squitti is author of more than 100 peer-reviewed publications. She is Scientific editor of Current Alzheimer Research, PLOS one. She greatly contributed to the understanding of Alzheimer’s disease (AD) by demonstrating the existence of a copper imbalance in AD patients. She was the first to discover a link between the clinical picture of AD and the existence of a restricted pool of copper that travels through the body loosely bound to and constantly exchanged among albumin and other low-molecular-weight compounds, i.e., differently from the bulk of copper which travels tightly bound to ceruloplasmin. The copper imbalance in AD consists in increased systemic redox-active non-Cp-Cu, decreased total copper in the brain and increased redox-active labile copper in the brain and resembles (although it is much less severe) the one characterizing Wilson’s disease, the paradigmatic disease of non-Cp-Cu accumulation and intoxication. In Wilson’s disease, mutations in the ATP7B, the gene that codes the pump controlling copper excretion through the bile, cause defective pumping in the hepatocytes, which, in turn, causes the release of non-Cp-Cu in general circulation. Evidence collected by Dr. Squitti’s group has demonstrated that abnormal non-Cp-Cu levels correlate with the clinical and anatomical deficits of the AD, with Aβ and Tau in the cerebrospinal fluid, with a worse prognosis of the disease and with the mild cognitive impairment (MCI) condition. A 6-year longitudinal study has demonstrated the predictive value of non-Cp-Cu levels in the MCI conversion to full AD. A landmark of Dr. Squitti’s work was the demonstration that some variants in the copper gene ATP7B are susceptibility loci for AD and that specific variants of this gene are risk factors for copper metabolic alterations in AD patients, contrary to others, which are instead protective towards this copper imbalance. This copper metabolic ‘subtype’ of AD affects about 60% of AD sporadic patients, who are typified by abnormal levels on non-Cp-Cu. A series of Dr. Squitti’s recent studies demonstrated peculiar neurophysiological, neuroimaging, as well as biochemical features, which distinguish the AD copper. The potential benefits appear enormous in terms of personalized medicine based on biomarker science.
