Bio:
Professor Patrick Humbert, BSc (Hons) University of Western Australia, PhD Walter and Eliza Hall Institute (University of Melbourne). In 1996, Dr Humbert undertook postdoctoral training at the Massachusetts Institute of Technology (MIT) where he made seminal contributions to understanding the mechanism of tumour proliferation. In 2000, Dr Humbert has set up his independent research laboratory at the Peter MacCallum Cancer Centre, Melbourne, and in 2016 relocated to La Trobe Institute for Molecular Science (LIMS) to help lead the Cancer Biology theme. His current focus is the study of how cell shape and orientation regulate stem cell function, organ formation and cancer. Professor Humbert’s laboratory has made major inroads in understanding how the genetic program that controls cell orientation can prevent the development of tumours of the breast and prostate. In 2001, Professor Humbert was awarded a Special Fellowship from the Leukemia and Lymphoma Society of America and is the recipient of consecutive Career Development Fellowships and now Senior Research Fellowship from the National Health and Medical Research Council of Australia.
Lab Overview:
Our laboratory has developed a program of research focused on understanding how maintenance of the architecture of tissue and the polarity of cells within a tissue regulates cell number and behaviour in vivo. We seek to utilise information obtained from Drosophila genetic screens and our own mammalian functional screens to apply to mammalian epithelial biology and to the study of human cancer. Our laboratory’s initial studies have determined that human homologues of Drosophila polarity genes such as Scribble operate similarly in mammalian epithelial systems to prevent tumour formation. We are examining how polarity proteins such as Scribble regulate tumourigenesis and establishing how their disruption in human cancer drives poor prognosis in cancer patients. In addition, we are utilising Drosophila as a platform for drug discovery together with structural studies of mammalian polarity regulators to develop therapeutics to target disease of tissue disorganisation such as cancer. A better molecular understanding of how tissue architecture and polarity are disrupted in cancer will lead to the development of novel therapeutic strategies.
Research focus:
▪ Analysing cell biology and cell polarity protein function in 3D epithelial cell culture models.
▪ Generating and analysing of Genetically Engineered Mouse Models (GEMMs) to investigate role of polarity proteins in cancer progression and organ development in vivo.
▪ Analysing cell polarity proteins and gene alterations in human cancers.
▪ Conducting large-scale RNAi functional screens to identify effectors of Cell Polarity.
