PhD PERIOD
From 1998 to 2004, I worked in the Genetics and Molecular Biology of mitochondrial proteins and related diseases Group in the Biochemistry Department (Faculty of Biology, University of Barcelona), under the supervision of Dr. Villarroya and Dr. Solanes, supported by a UB grant (University of Barcelona). I was working on the mechanisms of UCP2 and UCP3 (Uncoupling protein 2 and 3) transcriptional regulation. UCPs are proteins whose malfunction has been mainly related to obesity and diabetes, but also they have been involved in ROS generation and cancer. I achieved the PhD degree in 2004 with 7 international publications coming from this period (Pedraza, 2000, Solanes, 2000, Barbera, 2001, Pedraza, 2001, Solanes, 2003, Solanes 2005, Pedraza 2006). I was awarded a short-term stay grant in Dr. Vidal-Puig’s laboratory (Department of Clinical Biochemistry and Medicine, Addenbrooke’s Hospital, University of Cambridge) in 2003, where I learned some molecular biology techniques.
POST-DOCTORAL PERIOD
During my first postdoctoral period in Lleida (2005-2010, Faculty of Medicine, University of Lleida) I worked in the Cell Cycle Group, a broad team focused in different topics related to cell proliferation, differentiation and oncogenesis. In this period, I was receiving economical support by a Juan de la Cierva contract from Ministry of Science (2006-2009), and directed by Dr. Gallego, I was involved in two different projects related to protein kinases. Regarding the first project, I analyzed the effects of MLK2 (mixed lineage kinase2), a MAP3K involved in apoptosis and survival, in relation to proliferation and neuronal differentiation. MLKs are therapeutic targets in neurodegenerative diseases (i.e. Parkinson disease). In relation with the second project I described the function of KIS in local translation in neurons, a kinase related to schizophrenia, previously involved in cell cycle progression, which is overexpressed in leukemia cells. All together, this work has derived in two published papers (Cambray, 2009, Pedraza, 2009) and one Doctoral Thesis (M. Rafel, 2012) co-directed by me. In 2010 I moved to Barcelona (IBMB, CSIC) with Dr. Gallego, where I continued studying the mechanism by which KIS would enhance local translation in neurons and determining its functional relevance under normal and pathological conditions, such as schizophrenia (Pedraza, 2014, Ortiz, 2017). In 2014 I returned to the Institut de Recerca Biomèdica de Lleida (IRB Lleida) at the University of Lleida where I joined Dr. Eloi Gari’s group and I am now involved in a challenging project to elucidate the non-canonical functions of CyclinD1 in the cytoplasm. CyclinD1 is a proto-oncogen amplified in many cancers and accumulated in the nucleus of tumor cells. However, CyclinD1 also targets cytoplasmic proteins involved in the regulation of cell migration and invasion (Fuste, 2016, Fuste, 2016b, Cemeli, 2019, Fuste, 2019). Specifically, I am interested in revealing new functions of cytoplasmic CyclinD1 in neurons and I have supervised the Doctoral Thesis of M.Ventura Monserrat (2019), about the effects of CyclinD1 on GABA-A receptors. Nowadays, I am co-directing the Doctoral Thesis of M.Guasch, that deals with the role of CyclinD1/Cdk4 complex on cell signaling.
