Professor Howells is a stroke biologist who pioneered the pre-clinical use of systematic review and meta-analysis which together with laboratory experimentation has contributed to our understanding of the translational road blocks in stroke medicine and the evidence-based design of clinical trials of hypothermia in stroke (EuroHyp-1) and spinal cord injury (ICED, Batchelor et al, NHMRC Project Grant 1066217). Professor Howells began his research career studying vitamin A, E and D, developing methods for high throughput analysis and management of patient selection and sample collection in the difficult circumstances of a busy neonatal intensive care unit at St George’s Hospital in London. Even though still a research assistant, he was first author on all the key publications. His Ph.D., which resulted in 15 publications, developed methods to detect highly unstable cofactors and metabolites of neurotransmitter synthesis in the cerebrospinal fluid of very sick children at Great Ormond Street Hospital in London. These methods were applied to diagnosis of inborn errors of dopamine and serotonin metabolism and of CNS inflammation in children with white blood cell abnormalities and parasitic, bacterial and viral infections of the brain. As a post-doctoral fellow at the Murdoch Institute he identified the mutations which cause phenylketonuria and dihydropteridine reductase deficiency and while running the diagnostic enzymology laboratory developed methods to detect other inborn errors of metabolism. At the Austin Hospital he discovered a new population of dopaminergic neurons in the human striatum; showed that a BDNF deficit can cause Parkinsonism in rats and that such a deficit exists in Parkinson’s disease; discovered that macrophages and microglia form intimate contacts with regenerating dopaminergic axons and promote axonal outgrowth but provided surrogate targets that limit neuronal re-connection. More recently, he has established Australia’s most productive preclinical stroke research laboratory, a CSIRO stroke biomarker discovery program using animal models of stroke to fast-track human biomarker discovery; pioneered the use of systematic review and meta-analysis in the basic sciences; co-founded the multinational and multidisciplinary CAMARADES and Multi-PART collaborations to provide the best preclinical evidence to support stroke clinical trials and developed the use of human stem cell derived brain cells to probe human stroke biology.
