Mentored by Dr. Dan S. Kaufman, who is the first researcher to differentiate human embryonic stem cells (hESCs) into hematopoietic stem cells (HSCs) and a leader in human induced pluripotent stem cells (hiPSCs) derived Natural Killer (NK) cells, I studied the in vitro development of HSCs and lymphocytes (B cells, T cells, and NK cells) using hESCs/hiPSCs as a novel cell source and obtained a Master of Science (M.S.) degree. Combining gene editing techniques through collaboration with two institutes, two genetic X-linked immunodeficiency diseases were successfully modeled, gene-corrected, and differentiated into functional lymphocytes (T and NK cells). One is the severe combined immunodeficiency disease (X-SCID) caused by mutations in the IL2RG gene, and the other is Wiskott-Aldrich syndrome (WAS) caused by WAS gene mutation. I published one master's thesis (Title: Derivation of lymphocytes from human induced pluripotent stem cells. Advisor: Dan S. Kaufman) and co-authored four research articles.
Mentored by Dr. Paula P. Bickford and Dr. Daniel C. Lee, my doctoral research focuses on the roles of arginine metabolism and sensing in the neuroinflammation and neuropathology of Alzheimer’s disease (AD). One project studied the impact of macrophage/microglia Arginase 1 (Arg1) insufficiency during amyloidosis in the APP Tg2576 mice. Another project studied if manipulating the gene expression of a putative extracellular arginine sensor Gprc6a through recombinant adeno-associated virus (rAAV) vectors affect tau pathology in the PS19 mice. During my Ph.D., I have co-authored eight publications (two first authorship), written 15 conference abstracts, given 13 poster and two oral presentations. I have been awarded two outstanding poster competition awards, four travel awards, including the Christopher Phelps Memorial Fund travel award for the top graduate student in Neuroscience, and a prestigious Dorothy Benjamin Graduate Fellowship in Alzheimer’s Disease.
