My research interest in immunology, in particular in antigen processing and presentation, dates back to the early 1980’es. As a young medical doctor, I started my scientific career under the guidance of Dr. Ole Werdelin and became involved in demonstrating how two peptides compete for antigen presentation thereby providing the first functional evidence of specific oligopeptide binding to MHC class II molecules. Soon after, now working at National Jewish Center in Denver under the guidance of Dr Howard M Grey, I managed to demonstrate peptide-MHC class II binding biochemically. Creating my own laboratory in Denmark, I initiated a systematic analysis of human MHC class I and II molecules (most distinctly presented in two review papers proposing a “human MHC project”). To support this kind of large-scale project, I started systematically assembling the necessary tools: generating recombinant MHC class I and II molecules, generating high-throughput peptide-MHC interaction assays, generating peptide-MHC binding data, generating bioinformatics predictors, generating an iterative bioinformatics-biochemistry process, generating novel bioinformatics tools handing essential steps in T cell epitope discovery, generating a large-scale strategy for peptide-MHC tetramer production, and most recently, I have been the coordinator of an EU supported project successfully generating a high-density peptide microarray technology. Specifically, I am a partner in the NIH sponsored IEDB core project and PI on two consecutive large-scale T cell epitope discovery contracts. In brief, my research interests include: Immunology (antigen processing and presentation, MHC production and specificity, T cell specificity and activation, vaccination and immunotherapy), Biotechnology (cloning and sequencing, recombinant protein production, refolding and purification), Bioinformatics (databases, predictors). My administrative experiences include being coordinator/PI of four international European Union (EU) grants and PI of two NIH contracts. I have been partner on several additional other international projects (EU, NIH, NSF, Gates). In the two most recent EU grants, I have been coordinating the successful development of a high-density peptide microarray technology capable of representing the entire human proteome on one array.
