Scientific goals:
To advance our knowledge of polarized membrane trafficking through the functional characterization of the protein machinery involved in this process. Special emphasis is placed on the functional characterization of the specialized membrane trafficking pathways of polarized epithelial cells and T lymphocytes that are mediated by members of the MAL protein family and INF2, a MAL and MAL2 -interacting formin protein. Since 2013, the focus of our research is to investigating the biogenesis of the primary cilium of epithelial cells and analyzing the function of INF2, whose mutation causes focal segmental glomerulosclerosis and Charcot-Marie-Tooth neuropathy, in health and disease.
Main achievements (2000-present):
- Identification and functional characterization of MAL as a component of the specialized machinery of transport of newly synthesized proteins to the apical surface directly from the Golgi in polarized epithelial cells, and of a specialized route of apical endocytosis that is restricted to specific cell surface receptors (2000-2004).
- Identification and functional characterization of MAL2 as a central element in the indirect pathway of protein transport from the basolateral to the apical surface (transcytosis), which is necessary for the exit of cargo molecules from basolateral endosomes for sorting to the apical subdomain (2002-2006).
- Discovery of the MARVEL domain-containing superfamily of proteins (2003).
- Identification and functional characterization of an essential role for MAL in targeting Lck to the plasma membrane of T lymphocytes, for the formation of the immunological synapse and for T-cell receptor signaling (2007-2012).
- Functional characterization of INF2, a MAL- and MAL2-associated formin involved in membrane trafficking both in epithelial cells and T lymphocytes. Our group also participated in the characterization of INF2 mutations, which are responsible for Charcot-Marie-Tooth disease with associated focal segmental glorumerosclerosis (2007-2012).
- The involvement of formin INF2 in posttranslational modifications of microtubules by T lymphocytes and epithelial cells (2012-present)
- Proposal of a novel mechanism of primary cilium formation by polarized epithelial cells (2016-present).
