Main focus of my research is studying neuropathology, neurodegeneration and neuroinflammation using Positron emission tomography (PET) in Alzheimer's disease (AD). My first years in PET research included the world first studies of in vivo amyloid PET imaging in mild cognitive impairment (MCI) (Forsberg, et al, 2007) and longitudinal changes of amyloid load in AD (Engler, et al, 2006). I have after that continued AD research at the Karolinska PET center in Solna including several projects focusing on evaluation and application of novel PET radioligands targeting amyloid and neuroinflammatory targets such as the Translocator protein (TSPO) in microglia. One ongoing project involves investigation of patterns involving amyloid deposition in brain measured with [11C]AZD2184 PET and cognitive function. In a related study we investigated very early cognitive changes in healthy elderly subject and relate any changes to presence or absence of amyloid pathology. We could not find any elevated amyloid load in subjects with subtle cognitive problems, however there was connection between amyloid load and ApoE-ε4 genotype (Mattsson, et al, 2015).
I am also heavily involved in research with radioligands measuring inflammation in brain. We have for the last 5 years been heavily invested in evaluating and applying the TSPO radioligand [11C]PBR28. In a test/retest study we were able to determine that [11C]PBR28 shows medium reproducibility and high reliability in brain (Collste, et al, 2015). We have then applied [11C]PBR28 in various diseases and disorders including Psychosis (Collste, et al, 2017), Allergy (Tamm et al, 2017), Patients undergoing abdominal surgery (Forsberg, et al, 2017), (Chronic fatigue syndrome (SCFS) (Forsberg et al, manuscript), Rheumatoid Arthritis (Forsberg, et al, manuscript) and Fibromyalgia (Forsberg, et al, manuscript).
In AD I am currently involved in projects both regarding novel biomarkers for pathological entities such as amyloid plaques and Tau as well as targets to study neuroinflammation. On top of that the we are working closely with industrial partners in a number of studies focusing on novel disease modifying treatment strategies towards amyloid production and clearance as well as neuroinflammation. Our scope of research over the years ranges from evaluating the effect of symptomatic treatments of the Acetylcholine esterase inhibitors Rivastigmine (Stefanova, et al, 2006) and Galantamine (Keller, et al, 2011), Phenserine targeting amyloid beta production (Kadir, et al, 2008) and an occupancy study using the novel MAO-B inhibitor sembragiline, (Sturm, Forsberg, et al, 2017).
Other PET imaging projects I have been and are involved in includes studies of pathological changes in Parkinson’s disease investigating the serotonergic (Varrone, et al, 2014; Varrone, et al, 2015) and dopaminergic systems (Fazio, et al, 2015); investigation of inflammation and the serotonergic system in CFS in monozygotic twins (Takahashi, Watanabe, et al, manuscript), evaluating the serotonergic system in depression (Tiger, et al, 2014), and the dopamine system in anxiety (Plavén-Sigray, et al, 2017).
I am also involved in methodological development using new techniques for image and data processing. We have also published methodological papers on novel ways of quantifying [11C]PBR28 (Kanegawa, et al, 2016) and scrutinizing the use of simplified ratio methods for [11C]PBR28 quantification (Matheson, et al, 2017).
