RESEARCH SUMMARY
Dr. Ji has pioneered structural analysis of double-stranded RNA (dsRNA) in complex with selected members of the ribonuclease III (RNase III) family. During the decade of the 2000s, his bacterial RNase III structures provided the first structural view of dsRNA processing while establishing a two-magnesium(II)-ion catalytic mechanism. Over the last decade, his yeast Rnt1p structures revealed a double-ruler mechanism for substrate selection. All RNase III enzymes follow the two-magnesium(II)-ion mechanism of catalysis, but eukaryotes employ two more catalytic side chains for the cleavage of each phosphodiester bond. Dr. Ji’s structures of a post-cleavage complex determined at a stage immediately after the cleavage of scissile bonds for both prokaryotic and eukaryotic RNase III enzymes provided basis for meaningfully modeling the catalytic assemblies of other RNase IIIs such as Drosha and Dicer.
