From the beginning of my scientific training, I was very interested in addressing the importance of bacterial toxins in human pathogenesis. During my studies, I performed functional analyses on the interaction of several bacterial toxins, such as diphtheria, pertussis and anthrax toxins, with membranes, and how these toxins gain access to cells and participate in pathogenesis. However, the main focus of my PhD was the mechanism of action of clostridial neurotoxins. I demonstrated that the inhibition of synaptic activity caused by tetanus and botulinum neurotoxins is due to a specific protease activity directed towards three synaptic proteins playing fundamental roles in neurotransmitter release. This discovery was instrumental in the field of SNARE biology and provided the physiological validation for the involvement of synaptic SNARE proteins in neurotransmitter release, thus enabling the widespread exploitation of botulinum neurotoxins ad their derivatives in human therapy.
