The human body tightly sequesters essential micronutrients such as iron, zinc and manganese, restricting their access to invading microorganisms. This process is known as nutritional immunity. Therefore, microorganisms that cause disease must have developed specialised micronutrient uptake systems in order to grow in their hosts. I will establish how the major human fungal pathogen, Candida albicans, overcomes nutritional immunity during infection. I will use a powerful combination of genome-wid e profiling, genetics and molecular dissection to discover the mechanisms that C. albicans uses to acquire micronutrients. I will then employ specialised infection models to uncover exactly how nutritional immunity impacts the growth of C. albicans, and which mechanisms the fungus uses to overcome this restriction. Finally, I will determine how conserved these mechanisms are amongst other pathogenic fungal species. This will validate whether micronutrient uptake represents a broad-spectrum thera peutic target.
