(A) Kinase screening assays were conducted by Millipore's KinaseProfiler according to their established protocols. Scores represent the percent of control, which was derived from the following formula: % of control = [(sample − mean no enzyme)/(mean plus enzyme − mean no enzyme)] × 100. The kaempferol concentration for each specific kinase assay was set at 25 μM and ATP concentration was set at 10 μM. Data are representative of results from triplicate experiments. (B) Kaempferol can bind at the ATP binding pocket of RSK2. Enlarged view shows kaempferol binding with RSK2 at the ATP binding pocket and forming hydrogen bonds (shown as red lines). (C) Kaempferol can bind at the ATP binding pocket of MSK1. Enlarged view shows kaempferol binding with MSK1 at the ATP binding pocket and forming hydrogen bonds (shown as red lines). (D) Kaempferol does not bind with JNK2 or p38. Whole lysates from JB6 P+ cells were incubated with 30 μL of kaempferol-Sepharose 4B beads (50% slurry) for 2 h at 4{degree sign}C. Beads were washed and JNK2 or p38 binding was visualized by Western blotting.
