Establishment of a Human Breast Cancer Cell Line MDA-MB-231BM3 with High Bone Metastatic Potency and Its Animal Model in Immune Deficient Mice

Objective:To establish a human breast cancer cell line (MDA-MB-231BM3) with high bone metastatic potency and its animal model in immune deficient mice.Methods:The human breast cancer cells MDA-MB-231T were injected into the left cardiac ventricle of nude mice. The metastatic lesions of tumor-bearing mice were imaged by gamma camera. The mice were then sacrificed and the lesions of bone tissues were resected. Metastatic cancer cells in the resected lesions were subjected for culture. This process was repeated for 3 cycles to obtain a highly metastatic cell subline (MDA-MB-231BM3) and its animal model. MDA-MB-231BM3 and MDA-MB-231BO established in foreign countries were compared on the animal model of bone metastasis of breast cancer. Results:The study,successfully establishes MDA-MB-231BM3 cell line,which mainly migrates into bone tissues and sometimes migrates into lung and adrenal gland tissues,etc,and its animal model with bone metastasis in nude mice. Conclusion:MDA-MB-231BM3 has a high bone metastatic potential. The cell line and its animal model provide a good platform for research of the biological behaviors of metastasis of breast cancer.
