Conserved region amplified polymorphism(CoRAP) is a novel gene targeted marker technique based on expressed sequence tags with the advantages of simplicity,reproducibility and specificity.In this technique,the fixed primer is designed on the basis of expressed sequence tags that represents important agronomic traits,and random primers are designed according to the partial intrinsic conserved sequence of most of introns,and then dominant polymorphic markers with preferential targeting regions flanking expressed sequence tags can be generated and used to design the fixed primer for performing PCR amplification using the standard PCR procedure at annealing temperature of 52℃.The CoRAP molecular marker technique can be used as an alternative to conventional SRAP and TRAP techniques,and it can be widely applied in the fields of biodiversity analysis,genetic map construction,important agronomic traits gene tagging,QTL mapping and marker assisted selection breeding.
