Optimasi Metode Polymerase Chain Reaction (PCR) Untuk Identifikasi Polimorfisme Gen SLCO1B1 Sebagai Farmakogen Statin

Farmakogenomik merupakan suatu ilmu dengan menggunakan informasi genetik yang bertujuan untuk mengurangi efek obat yang merugikan. Informasi genetik yang digunakan biasanya dalam bentuk Single Nucleotide Polymorphisms (SNP), salah satunya yaitu SLCO1B1. SLCO1B1 merupakan salah satu variasi genetik yang dapat mempengaruhi toksisitas suatu obat, salah satunya yaitu statin. Statin dikenal sebagai obat yang memiliki efek samping berupa miopati. Varian alel SLCO1B1 yang sering dikaitkan dengan statin adalah rs4149056 atau SLCO1B1*5. Penelitian ini bertujuan untuk membuat primer yang spesifik untuk mengidentifikasi polimorfisme gen SLCO1B1 (rs4149056) dan menetapkan kondisi optimum dari metode PCR yang sesuai pada identifikasi polimorfisme gen SLCO1B1 dalam statin. Penelitian dilakukan secara in silico dan lab basah di Prodia Jakarta. Hasil penelitian menunjukkan bahwa primer forward 5’-GGT TGT TTA AAG GAA TCT GGG TCA T-3’ dan primer reverse 5’-GCA GCA GCC ACA AGA AGA CT-3’ yang telah didesain mampu mengidentifikasi polimorfisme gen SLCO1B1 (rs4149056) pada kondisi optimal konsentrasi DNA template 10 ng/5 μl dan suhu annealing 59°C. Sehingga, dapat disimpulkan bahwa polimorfisme gen SLCO1B1 (rs4149056) berpengaruh sebagai farmakogen statin. Pharmacogenomics is a science using genetic information that aims to reduce the adverse effects of drugs. The genetic information used is usually in the form of Single Nucleotide Polymorphisms (SNPs), one of which is SLCO1B1. SLCO1B1 is one of the genetic variations that can affect the toxicity of a drug, one of which is statins. Statins are known as drugs that have side effects in the form of myopathy. The SLCO1B1 allele variant often associated with statins is rs4149056 or SLCO1B1*5. This study aims to create a specific primer to identify the SLCO1B1 (rs4149056) gene polymorphism and establish the optimum conditions of the appropriate PCR method on the identification of SLCO1B1 gene polymorphisms in statins. The research was conducted in silico and wet lab at Prodia Jakarta. The results showed that the forward primer 5'-GGT TGT TTA AAG GAA TCT GGG TCA T-3' and the reverse primer 5'-GCA GCA GCC ACA AGA AGA CT-3' that had been designed were able to identify SLCO1B1 gene polymorphism (rs4149056) under optimal conditions of template DNA concentration of 10 ng/5 μl and annealing temperature of 59 °C. Thus, it can be concluded that the polymorphism of the SLCO1B1 gene (rs4149056) has an effect as a statin pharmacogen.
