•ECL sensing platform with programmable entropy-driven cycling strategy and inverted DNA tetrahedral nanomachines.•Ti3C2Tx nanomaterial introduced into ECL sensing platform for SARS-CoV-2 RdRp gene detection.•ECL platform with wide range of 100 aM to 10 nM and LOD of 48.3 aM.•ECL sensing platform with recovery rates of 97% to 103.3% and 98.34% to 103% in real/clinical settings.
