•SGLT2 expression and O-GlcNAcylation was increased in human NASH liver.•SGLT2 expression correlated with the degree of hepatic fibrosis in humans.•Upregulation of SGLT2 increased O-GlcNAcylation in hepatocytes under NASH condition.•SGLT2 inhibition improved NASH phenotypes via modulating O-GlcNAcylation and AMPK.•Reduced O-GlcNAc by SGLT2 inhibition enhanced AMPK-TFEB mediated autophagic flux.
