Isolation and Evaluation of Antioxidants from Arisaema Heterophyllum Tubers

To the best of our knowledge, we, for the first time, isolated eleven compounds from the dried tubers of Arisaema heterophyllum, including two steroids (1 and 2), one triterpenoid (3), two flavonoids (4 and 5), one lignan (6), and five phenyl derivatives (7-11). Compound structures were established by performing detailed analysis of nuclear magnetic resonance (NMR), the structure of 1 was determined using high-resolution electrospray by ionization mass spectrometry and NMR. Absolute configurations of rhamnose and glucose in compounds 5 and 10 were determined using optical rotation. Compounds 7 and 9 were first discovered in natural products. Six key oxidation targets were screened based on network pharmacology, and their possible biological signaling path-ways were predicted by Gene ontology analysis. Molecular docking was employed to analyze interactions be-tween compounds 1-11 and six key targets. Antioxidant activities of compounds 1, 4, 6-9, and 11 were evaluated using DPPH radical-scavenging experiments. Compounds 1, 4, and 8 exhibited notable antioxidant activities at 50 mu M, with %inhibition values of 36.29 +/- 0.02, 33.14 +/- 0.02, and 20.35 +/- 0.01 %, respectively, L -ascorbic acid was used as the positive control (65.93 +/- 0.01 %). Based on molecular docking, the antioxidant effects of examined compounds may be related to the target proteins SLC6A4, CASP3, and BAX.
