•PKG regulation of RyR2 has been under-researched relative to other kinases.•Endogenous PKG activation reduced the threshold for spontaneous RyR2 Ca2+ release.•Regulation of RyR2 by PKG required expression of serine residues 2808 and 2030.•Knockdown of PKG found minimal basal regulation of RyR2 by PKG.
