Event Abstract Back to Event B lymphocytes activation in GALT (Gut Associated Lymphatic Tissue) in immunized hens with Escherichia coli K88. These come in contact with Mucosal Immune System (MIS), specifically the portion that relates to the digestive tract (Gut Associated Lymphoid Tissue, GALT). Here, avian intestinal lymphocytes, inducing a specific response to the pathogen by producing IgY that is deposited in the egg yolk to provide passive immunity to the future chicks. Once the egg is laid, the specific IgY is easily removable by different methods and act as immunoprophylactic and/or immunotherapeutic ways, in different enteric diseases such as diarrheas in piglets caused by Escherichia coli K88 (E. coli K88). The objective of this work was to study humoral immune response in GALT hens, determining germinal centers response post immunization with Escherichia coli K88 lysate. Thirty four (Shaver) hens were divided in two groups: K88 (n = 25) and Control (n = 19). At 22 weeks old, the hens were immunized with a E. coli K88 bacterin, using the E coli most prevalent in pig farms in our region, formaldehyde inactivated E. coli K88, 1x109 UFC/bacteria) or adjuvant (aluminum hydroxide), 0.5 ml in each pectoral muscle. Four immunizations were carry out every two weeks After each immunization, blood samples were taken and were used to determine the concentration of total and specific IgY and IgA by ELISA test. Eggs were collected during 7 weeks and separated by water soluble fraction containing IgY. At 44 weeks old, hens were killed by cervical dislocation and blood samples were taken for quantify B cells by flow cytometry. Then were take off: esophageal, pyloric and cecal tonsils, spleen and gut samples which were processed and stained by conventional and immunofluorescent histological technique. Histomorphometry of the villi and crypts area Lieberkühn and the area of the germinal centers activated in GALT and spleen was measured by Axio Vision Release and Image J programs. A portion of spleen was used to quantify B cells by flow cytometry. In addition, secretory IgA concentration was measured in gut mucus by ELISA test. Data were analysed by ANOVA and Turkey´s test using Infostat program, p≤ 0.05 were considered significant. Result: K88 hens had higher numbers of peripheral B cells and spleen than Control group (p ≤ 0.05). Also, K88 group showed higher IgA serum levels (p≤0.05), higher total and specific IgY serum levels (p≤0.001), higher secretory IgA concentration in gut mucosae (p≤0.001) and higher IgY concentration serum and in egg (p≤0.001) than Control groups. In addition, the bacteriae immunized group presented more active germinal centers, with abundant B lymphocytes in esophageal Tonsils, Peyer's patches (p≤0,001) and pyloric and cecal tonsils (GALT) (p ≤ 0.05) and spleen (p≤0,001). Abundant fluorescent intraepithelial B cells and in propria laminae gut were detected. In histomorphometric gut study, K88 group had taller villi (p≤0.05) than Control group. Increased epithelial cells number intermixed with plasma cells was noticed in bacterin´s immunized hens and smaller globelet cells number than Control in villi. The Lieberkühn zone cryptae was taller in K88 group (p ≤ 0.05) than Control. It be concluded that the immunization of hens with Escherichia coli K88 lysate, IM, activated by one hand mucosal protection, producing high IgA levels and in another hand systemic protection producing high specific IgY levels.
