摘要
Elevated expression of insulin-like growth factor-II (IGF-II) is a frequent genetic abnormality seen in hepatocellular carcinoma (HCC). For a better understanding of its role in maintaining the malignant phenotype, we examined the anti proliferation and anti-anoikis effects elicited by a decrease in the transcription and protein levels of IGF-II by RNAi in HepG2 cells. Real-time qPCR analysis shown that transfection of siRNA targeting IGF-II was 85.3% at mRNA and 75% at protein levels. Transfected with miRNA-IGF-II significantly decreased cell proliferation (P<0. 05) and rendered cells sensitive to anoikis (P < 0. 01), Indicating that the expression of IGF-II is associate with proliferation and the transformed phenotype of HCC cells in vitro, suggesting a promising strategy for targeted therapy in HCC and forming a basis for future in vivo trials.
