类风湿关节炎成纤维样滑膜细胞背景特异性microRNA-靶基因及microRNA-小分子互作网络分析

Abstract
目的 建立类风湿关节炎成纤维样滑膜细胞(rheumatoid arthritis fibroblast like synoviocytes,RA-FLS)背景特异性的microRNA-靶基因和microRNA-小分子互作网络.方法 利用microRNA PCRArray数据(GSE72564)筛选出RA-FLS和骨关节炎对照样本中差异表达的microRNAs,整合多个microRNA-靶基因互作数据库,构建RA-FLS差异表达microRNA与靶基因的功能调控网络;整合microRNA-小分子互作数据,构建RA-FLS差异表达microRNA-小分子调控网络,计算网络中心性,获得核心调控micmRNA及小分子药物节点.结果 获得RA-FLS差异表达microRNA 63个,建立RA-FLS背景特异性的microRNA-靶基因和microRNA-小分子互作网络,筛选出hsa-let-7b-5p、hsa-mir-15b-5p、hsa-mir-218-5p、hsa-mir-30a-5p、hsa-mir-520d-3p五个关键microRNA,5-氟尿嘧啶(5-fluorouracil)、glucose、1,2,6-Tri-O-galloyl-beta-D-glucopyranose (TGGP)、糖皮质激素(glucocorticoid)四种与RA-FLS密切相关的活性小分子.结论 RA-FLS背景特异性的microRNA-靶基因和microRNA-小分子互作网络的构建,尤其是中心性microRNA和小分子的筛选,有望成为RA-FLS研究的潜在靶点.
