Abstract
目的 建立超高效液相色谱-串联质谱法测定淡水鱼中孔雀石绿、结晶紫及其代谢物残留的方法.方法 采用对样品用乙腈提取,中性氧化铝固相萃取小柱净化后,超高效液相色谱-串联质谱测定,氘代孔雀石绿为内标物定量.色谱柱:AQUITY UPLC BEH C18(2.1mm×50mm,1.7 μm);柱温:40℃;样品室温度:4℃;流动相:A相为乙腈,B相为5 mmol/L乙酸铵+0.1%甲酸水,梯度洗脱;流速:0.25 ml/min;进样体积:10μl;多反应监测(MRM)模式检测.结果 最低检出限孔雀石绿、结晶紫、隐形孔雀石绿、隐性结晶紫均为0.05 μg/kg.孔雀石绿、结晶紫、隐形孔雀石绿、隐性结晶紫的平均回收率和RSD分别为91.9%(RSD=1.9%)、96.2%(RSD =3.3%)、100.2%(RSD =4.6%)、89.5%(RSD =4.1%).结论 本方法检出限低,精密度高,适用于淡水鱼中孔雀石绿、结晶紫及其代谢物的检测.
