Abstract
目的:探讨miR-21低表达能否增强苦参碱( matrine,MAT)对肝癌细胞的促凋亡作用。方法:实时荧光定量PCR(RT-qPCR)检测不同浓度MAT作用HepG2细胞后miR-21的表达情况。流式细胞术检测miR-21低表达对MAT诱导的细胞凋亡的影响;RT-qPCR和Western blot检测miR-21低表达对MAT诱导的细胞凋亡中Bcl-2和Bax mRNA和蛋白表达水平的影响。结果:miR-21表达量随MAT作用浓度的增加而增加;miR-21低表达可促进MAT诱导的细胞凋亡,并促进Bax mRNA和蛋白的表达( P<0.05),而抑制Bcl-2 mRNA和蛋白的表达( P<0.05)。结论:miR-21低表达可通过抑制Bcl-2和促进Bax的表达来增强MAT诱导的HepG2细胞凋亡。
