山慈菇中总黄酮的提取工艺优化

Abstract
目的:建立山慈菇中总黄酮的含量测定方法,并优化其提取工艺.方法:采用紫外-可见分光光度法测定山慈菇中总黄酮的含量.以山慈菇中总黄酮含量为考察指标,以乙醇体积分数、料液比、提取时间、提取次数为考察因素,在单因素试验的基础上,采用L9(34)正交设计优化山慈菇中总黄酮的提取工艺.结果:芦丁检测质量浓度线性范围为8~48μg/mL(r=0.9997);定量限为0.54μg/mL,检测限为0.18μg/mL;精密度、稳定性、重复性试验的RSD均小于2%;加样回收率为99.61%~102.38%(RSD=1.15%,n=6).最优提取工艺为70%乙醇、料液比1:20(g/mL)、提取时间45 min、提取次数2次.在此条件下,山慈菇中总黄酮的平均含量为2.74 mg/g.结论:所建含量测定方法简单、准确;所得提取工艺稳定、可行.
