Micropropagation of Spathoglottis Eburnea Gagnep, a Thai Orchid Species, Through Shoot Tips

Spathoglottis eburnea Gagnep is a Thai terrestrial orchid species grown in the north and northeast. It has a small population in the wild and can be endangered in the near future. To develop a protocol for Spathoglottis eburnea micropropagation, shoot tips were proliferated in half strength Murashige and Skoog (1/2 MS) liquid media supplemented with the combination of a-naphthalene acetic acid (NAA) and 6-benzylaminopurine (BAP) (0, 0.5, 1 or 2 mg L-1) and placed on a shaker at 110 rpm. After 3 months of culture, the highest shoot formation of 30.8 shoots explant(-1) was obtained on 1/2 MS medium supplemented with 1 mg L-1 NAA and 2 mg L-1 BAP. Whereas, the combination of 2 mg L-1 NAA and 0.5 mg L-1 BAP gave the highest root formation of about 11.1 roots explant(-1). Moreover, the regenerated plants were analyzed by using flow cytometry and the results showed the same ploidy level.
