•Nigral and VTA rotenone infusion mediate sperm and testicular abnormalities.•The lesion compromised testicular tight junctions and germinative elements.•The brain lesion enhanced testicular caspase-3 but decreased SRY and GADD45 g signal.•The lesion caused testicular oxidative damage and reduced acetylcholinesterase activity.•The lesion was associated with reduced testosterone but FSH was not affected.
