•The role of endogenous sirtuin 6 in bone marrow-derived cells in atherosclerosis is unknown.•Loss of Sirt6 in bone marrow-derived cells is proatherogenic and increases macrophage scavenger receptor 1 expression.•Loss of Sirt6 in enhances oxLDL uptake in human and murine macrophages.•These findings assign endogenous SIRT6 in macrophages an atheroprotective role.
