•Propofol could prevent d-GalN/LPS-induced acute liver injury in mice.•The protective effects of propofol reflected in inhibition of inflammation, oxidative stress and hepatocyte apopotosis.•Propofol exerted its anti-inflammatory, anti-oxidative, and anti-apoptotic activity by regulating TLR4/NF-κB/NLRP3 pathway.•Propofol is promising to serve as a hepatoprotective agengt for the treatment of liver damage.
