•Matrine ameliorates CNS inflammation, demyelination, and astrogliosis in EAE.•Matrine decreases expression levels of SPH, S1P and SPHK1/2 in the CNS of EAE.•Matrine increases plasma gelsolin concentration in the CNS of EAE.•Matrine suppresses S1P1 expression in both CNS tissues and astrocytes of EAE.•Matrine inhibits activation of astrocytes and their SIP1 expression in vitro.
