Antimicrobial profile of Enterobacteriaceae and Vibrionaceae isolated from Cerastoderma edule.

Since cockles are filter feeders, they present the potential to accumulate great numbers of microorganisms including pathogens which can constitute a substantial food hazard when consumed raw or undercooked. Enterobacteria and autochthonous marine bacteria as Vibrionaceae present in this type of products may be very important due to its antimicrobial resistance. The aim of this study was to investigate the presence of antimicrobial resistance bacteria and its molecular determinants in cockle (Cerastoderma edule) harvested during November 2016 in Óbidos Lagoon, Óbidos, a shallow coastal lagoon in Portugal. Bacteria were isolated in selective medium for Enterobacteriaceae and Vibrionaceae containing the antimicrobials cefotaxime, ciprofloxacin and imipenem. Colonies with morphological distinct characteristics were selected from each media for further studies. The presence of genes qnrA, qnrB qnrS, qnrC, qnrD, aac(6’)-ib- cr, qepA, bla CTX-M , bla SHV , bla TEM , bla VIM , bla IMP-5 , bla KPC-2, bla NDM-1 , bla GES was assessed by PCR. Positive isolates to the presence of resistance genes were identified by 16sDNA PCR amplification and sequencing and the resistance genes alelles for bla TEM and bla CTX-M were determined the same way. Only some Enterobacteriaceae (n=6) were found positive for the presence of antimicrobial resistance determinants: qnrB (n=4), which confers low-level resistance to fluroquinolones; bla TEM-1 (n=1), an allele that only confers resistance to early cephalosporins in addition to many penicillins while other bla TEM alleles confer resistance to extended spectrum beta-lactams; and bla OXY (n=2), a chromosomally encoded beta-lactamase that confers resistance to aminopenicillins and carboxypenicillins, found when screening for the presence of bla CTX-M . The qnrB carriers were identified as Citrobacter spp, one Escherichia coli carried bla TEM-1 , one Klebsiella oxytoca isolate carried bla OXY while other carried both bla OXY and qnrB. The collection of isolates studied, despite having the presence of some resistance genes, did not reveal the presence of metallo-beta- lactamases or extended-spectrum- beta-lactamases since the genes found only confer resistance to early cephalosporins and penicillins and low-level resistance to fluorquinolones. The data shows that cockles from Lagoa de Óbidos do not present an excessive risk to the consumer with regards to infection with antimicrobial resistance bacteria.
