Iatrogenic Pneumopericardium and Cardiac Tamponade after Fiberoptic Bronchoscopy with Transbronchial Biopsy

1053 Objectives The aim of this study was to perform the cytotoxicity study of 188Re-human serum albumin microspheres (188Re-HSAM) in GP7TB cell line. The study was confirmed the inhibition effect of 188Re-HSAM in GP7TB hepatoma cell line. Methods HSA microspheres were synthesized by gradually-heating method. Amino borane-reduced 188ReO4− was interacted with carbon oxide to form [188Re(OH2)3 (CO) 3]+.[188Re(OH2)3(CO)3]+ was employed as a precursor for directly labeling HSA microspheres with 188ReO4−. Cytotoxicity assay was performed using 6-well disks. Individual assay of a 6-well disks were inoculated with 2 ml DMEM medium and 10%FBS containing 1×105 cells. All disks were incubated for 24 h to achieve 60-70% confluency. Administrated 188Re-HSAM and 188Re in serial dilution to each 6-well disks. The dose of 188Re-HSAM and 188Re was 0, 1, 10, 100 and 1000 μCi per assay. GP7TB cells were incubated with 188Re-HSAM and 188Re for 24 hour at 37 degrees. 24 hours later, the cells were washed twice with PBS, and dissolved in 10% trypsin. The numbers of the alive cells were calculated. Results The size of HSA microspheres was distributed between 20~35 μm. The yield of [188Re(CO)3(OH2)3]+ was 75~80%. The labeling efficiency of 188Re-HSAM was more than 85%. The viability of GP7TB cells was 100%, 87%, 81%, 39% and 21% with 188Re-HSAM in 0, 1, 10, 100 and 1000 μCi/assay, respectively. The viability of GP7TB cells was 100%, 101%, 91%, 66% and 30% with 188Re in 0, 1, 10, 100 and 1000 μCi/assay, respectively. The viability of GP7TB cells was 97% with HSAM.
