67Total FAK protein expression is related with anti-angiogenic therapy response in colorectal cancer models

Background: Focal adhesion kinase (FAK) is a multifunctional regulator of cell signaling, its expression has been reported in several tumors. Our group previously report FAK expression as predictor of tumor response in local rectal cancer. Its functional role in angiogenesis is not completely understood. Anti-VEGF therapy is frequently used in metastatic colorectal cancer. This is a preliminary report of possible FAK participation in colorectal cancer animaĺs models treated with standard anti-angiogenic therapy. Methods: We performed DLD-1 and HCT 116 animals models and treated with intraperitoneal bevacizumab and subcutaneous aflibercept every 3 days, for 21 days. Tumor preservation for RNA quantification, immunohistochemical (IHQ) analysis and protein expression was done. Functional analysis of pFAK (Tyr397) and total FAK protein expression in animal’s models were evaluated by western blot assay. Central tendency measure analysis and t student to compare median analysis were done, p value <0.05 was considered significant. Results: DLD-1 and HCT116 animals models had good response to anti-VEGF therapy. Significant decrease in total FAK protein expression was observed in both animaĺs models after anti-angiogenic therapy. An increase in FAK phosphorylation was also detected. Conclusions: This preliminary data suggest a decrease in total FAK after anti-VEGF therapy, these mechanism could be associated with good response to anti-angiogenesis therapy.
