Abstract
Objective To establish an HPLC methods for the simultaneous determination of human plasma concentration of glycyrrhizic acid and matrine.Methods Ultimate AQ-C18(250mm ×4.6mm,5μm)was chosen as the chromatographic column.The column temperature was at room temperature(25℃).The mobile phase was composed of acetonitrile and 0.05mol·L-1 aqueous phosphoric acid(22.5∶77.5,adjusted to pH 7.5with triethylamine).The flow rate was set at 1.0mL·min-1.The detection wavelengths were set at 220and 250nm.The injection volume was 50μL.Results The linear ranges of glycyrrhizic acid ammonium salt and matrine were 2.0-200.0μg·mL-1and 2.0-200.0μg·mL-1,respectively.The average recovery was above 95%; both the intraday RSD and the interday RSD were less than 4%.Conclusions The method is simple,rapid,sensitive and reliable, and could be used for the simultaneous determination of human plasma concentration of glycyrrhizic acid and matrine.
