•Metabolomics identified markers associated with high mannose (HM) glycan levels.•Ornithine was the top marker identified for HM level on secreted monoclonal antibody.•HM can be modulated by ornithine, spermine and copper levels in the media.•Correlation between HM and arginase 1 mRNA expression levels is observed.•Modulation of HM levels by polyamine pathway and cellular redox is proposed.
