Measurement of1J(Ni,C?i),1J(Ni,C?i?1),2J(Ni,C?i?1),2J(HNi,C?i?1) and2J(HNi,C?i) values in13C/15N-labeled proteins

Use of partial or selective C-13/N-15 labeling of specific amino acid residues in a given protein to measure the values of (1)J(N-15,C-13(i)alpha), (2)J(H-1(N),C-13(i)alpha), (2)J(N-15(i),C-13(i-1)alpha), (2)J(N-15(i),C-13(i-1)alpha) and (2)J(H-1(N),C-13(i-1)alpha) is described. This was achieved by recording a sensitivity-enhanced 2D [N-15-H-1] HSQC experiment, without mixing the spin states of C-alpha and C' during the course of entire experiment.
