The relationship between structure and function of membrane proteins, in particular transporters, has fascinated me since I began my scientific career. For a full understanding of a protein’s function, one always has to integrate the details into a larger picture by utilizing a multidisciplinary approach, which encapsulates a specialized set of tools required to answer specific biological questions. This integrative approach is routinely used in the field of Physiology as reflected in my biomedical sciences training.
During my undergraduate research education, I developed basic techniques in cell culture, molecular biology and protein biochemistry. Later, during my Ph.D., I gravitated towards structural biology of membrane proteins pursuing single particle electron microscopy on large respiratory chain supercomplexes. During these studies I appreciated that membrane proteins cannot be viewed without their lipid membrane context which can have significant influence on the functional and structural properties of the proteins. For further training in additional methods, I joined the lab of Eric Gouaux where I learned X-ray crystallography as another complementary technique for the structure determination of membrane proteins and to work with advanced eukaryotic expression systems. There I was also exposed to other biochemical and electrophysiological methods to elucidate the structure/function relationship of a ligand-gated ion channel again finding a close relationship between the protein and specific lipids. In the lab of Dr. Abramson I am currently employing my previously-acquired skills by establishing a mammalian expression system for transporters of the Sodium/Glucose Co-Transporter (SGLT) family. Furthermore, I am be able to gain deeper insights into lipid-based crystallization techniques, structure determination and additional biophysical methods for the study of membrane transporters. As project scientist I manage lab business, funds and equipment, contribute to grant proposals, mentor undergrad and graduate students and contribute to teaching.
