Upon the completion of PhD (2010) in which RC was dedicated to the synthesis of new heterocyclic compounds with potential biological activity, he has been focused in his research on the culture of animal cells (five years of post-doctoral experience).
In Mountain Research Centre (CIMO), the host institution, RC has been implementing various techniques using animal cells culture. His work was pioneer in the host institution, where RC supervised the implementation of the laboratory of cells culture and all the equipment existing there. Thus, it was possible to establish numerous bioassays that complemented the work of the research group (BioChemCore, http://esa.ipb.pt/biochemcore/).
Initially, RC implemented a screening system for the evaluation of the cytotoxic activity of natural extracts and synthetic compounds. He established five cultivated human tumor cell lines (HeLa, HepG2, MCF7, HCT15 and NCI-H460), in which hundreds of extracts/compounds were tested using the method of Sulforhodamine B.
Since it was needed to understand the degree of toxicity of the extracts/compounds under study, and knowing that primary cell cultures are very expensive, RC also implemented a method to obtain primary cell cultures from porcine liver. Thus, the research group also began to evaluate the hepatotoxicity of extracts/compounds. RC explored methods of co-culturing human tumor lines with human erythrocytes to assess the increased stiffness of the membrane of the erythrocytes in the presence of tumor cells.
RC established a method to evaluate the anti-inflammatory activity of extracts/compounds using mouse macrophages.
Currently, RC has been very dedicated to the application of techniques using flow cytometry to the animal cells culture. He is deploying and optimizing protocols to evaluate not only the apoptosis levels induced by extracts/compounds but also their interference in the cells cycle, trying to elucidate the possible mechanisms of action involved.
Over the past years RC has participated in several research projects funded by various institutions such as FCT and PRODER. His contribution is focused on the evaluation of cytotoxic effects. It should be highlighted that most of the projects are collaborations between the host institution and enterprises in the agro-food sector.
The collaboration with research groups from other countries (Spain, Serbia, Netherlands, Brazil, Egypt, Tunisia) is a major accomplishment of RC
RC has published in international scientific journals indexed to ISI (60 articles) most of them in Q1 (496 citations; H index 15). In addition, his research has also been presented in national and international conferences
RC was involved in the organization of the first workshop of animal cells culturing (2012), which was repeated every year until now. Throughout this time RC has also accompanied several MSc and PhD students, participated in master juris and receives regularly several manuscripts for peer review from journals of the SCI.
