The pharmacology of lower urinary tract organs has been the field of my career since I started my PhD studies. I started working with isolated urinary bladder from rats chronically treated with L-NAME, a nitric oxide synthase inhibitor. We have observed an overactive state of the bladders from these animals, leading us to conclude that the lower levels of nitric oxide at the level of urethra and prostate led to an overactive state on the bladder outlet, as the NO-cGMP pathway enzymes are more expressed in the urethra and prostate than in the bladder. During the years of my PhD, I also tested cGMP-increasing substances mainly sGC stimulators and activators in animal models of lower urinary tract tissues from hypertensive, diabetic, obese and cystitis animals. One of my main interests is to work with drug repurposing. For instance, my research group has published studies focusing on the role of mirabegron, a beta-3 adrenoceptor agonist approved by the regulatory agencies for treating patients with overactive bladder. Mirabegron relaxed prostate from rabbit and human in a mechanism dependent on beta-3 adrenoceptor activation. On the other hand, mirabegron relaxed corpus cavernosum from rat in a mechanism independently of beta-3 adrenoceptor stimulation. During 2014-2016 I did my post-doc under the supervision of Prof. Ferid Murad (Nobel Laureate in 1998) in the Department of Biochemistry and Molecular Medicine at George Washington University, Washington DC, USA. During this period, I investigated the role of epigenetic drugs in controlling the gene and protein levels of soluble guanylate cyclase in breast cancer cells. Since 2016, I have been studying the role of multidrug resistance proteins (MRP) in controlling cGMP or cAMP levels in lower urinary tract organs and in platelets in healthy and disease states.
