Platelet aggregation plays a central role in thrombosis and hemostasis, which are central to vascular disease. In the platelet lineage, the early committed cells undergo a unique cell cycle leading to the formation of polyploid megakaryocytes prior to their fragmentation into platelets. Our research focuses on mechanisms that regulate lineage commitment and the subsequent steps of cellular maturation of bone marrow cells into polyploid megakaryocytes. We reported that polyploidy also develops in aging vascular cells, and in other cell types under stress, and mechanisms of control of polyploidy and its relevance to disease states are investigated. An ongoing related project involves the molecular characterization of platelet and vascular adenosine receptors, and exploration of their role in cellular function and lineage development. The systems used include primary bone marrow cultures, as well as transgenic and knock out mouse models.
