A lo largo de más de 25 años, la actividad investigadora del grupo GENOXPHOS se ha centrado en el estudio de la cadena de transporte de electrones mitocondrial de mamíferos (MtETC) y la H+-ATP sintasa, que constituyen el sistema de fosforilación oxidativa (OxPhos). Entre sus aportes científicos se encuentran los estudios:
(1) Consecuencias funcionales de la variabilidad genética del mtDNA, siendo el trabajo más destacado el demostrar en humanos y ratones que la variabilidad poblacional del mtDNA condiciona el metabolismo del organismo, su respuesta a fármacos, predisposición a enfermedades, al envejecimiento saludable y ayudar a explicar la patología límite y la variabilidad funcional de las alteraciones del mtDNA. En conjunto, sus contribuciones destacan el papel de las ROS mitocondriales en la adaptación del sistema OxPhos a los requisitos metabólicos de la célula.
(2) Desarrollo de nuevos modelos de organización estructural del transporte mitocondrial electrónico. A partir de las observaciones y metodología desarrollada por el Dr. Schägger, los trabajos del grupo GENOXPHOS están transformando la comprensión de la estructura y función de la cadena respiratoria mitocondrial, dando lugar a la propuesta del "Modelo de Plasticidad" para explicar la organización dinámica de la cadena de transporte de electrones mitocondrial. Por un lado, este modelo, y el trabajo del que se deriva, explican el valor funcional de las asociaciones de complejos respiratorios en superestructuras, describe el primer factor proteico realmente necesario para la interacción física entre complejos. Demuestra la organización dinámica de la cadena respiratoria para optimizar el uso de diferentes fuentes de carbono y proporciona la prueba experimental del modelo de plasticidad propuesto. Su investigación ha permitido conectar la dinámica mitocondrial con la función bioenergética. Asimismo, en el contexto del modelo de plasticidad, se ha podido explicar el papel determinante de las isoformas estructurales del complejo IV de la cadena de transporte de electrones mitocondrial en su capacidad de homodimerización y en su capacidad de interactuar con otros complejos.
