My major research focus over the last 30 years has been the Glycoimmunobiology of the protozoan parasite, Trypanosoma cruzi, the causative agent of Chagas disease (CD). Particularly, I have been studying the structure, and biological and immunological roles of glycosylphosphatidylinositol (GPI)-anchored glycoconjugates (i.e., glycoproteins and glycolipids) that are major components of the parasite cell surface. Using the knowledge obtained over the years from these basic research studies, we are moving towards translational applications. For instance, more recently, we have been developing synthetic glycan- and peptide-based CD vaccines and biomarkers (BMKs) for the early assessment of chemotherapy outcomes in CD. In that regard, since August 2018, I have been the Program Director (PD)/Principal Investigator (PI) of the NIH-funded, phase-2b U01 clinical trial, entitled “TESEO” (https://clinicaltrials.gov/ct2/show/NCT03981523), for testing new chemotherapeutic regimens of the two existing drugs (benznidazole [BNZ] and nifurtimox [NFX]) and new BMKs for early assessment of therapeutic outcomes in chronic CD. Throughout my career, I have also been actively involved in the structural and functional analysis of a variety of biomolecules from different sources and organisms, including other parasites (Leishmania spp., Trypanosoma brucei, Plasmodium vivax), fungi (Cryptococcus neoformans, Histoplasma capsulatum, Paracoccidioides brasiliensis), bacteria, arthropod vectors (triatomines, ticks), and cancer cells, collaborating with several research groups in the U.S., Latin America, and Europe. Since, November 2004, I have been the director of the Biomolecule Analysis and Omics Unit (BAOU) (formerly, Biomolecule Analysis Core Facility), Border Biomedical Research Center (BBRC), UTEP, which is a cutting-edge core facility for omics (e.g., proteomics, lipidomics, and glycolipidomics). The Unit has an array of mass spectrometry (MS) instruments (i.e., ESI-Hybrid Quadrupole-Orbitrap-MS, MALDI-TOF-MS, GC-TSQ-MS, and ESI-TSQ-MS, ESI-LTQ-MS, ), IsoPlexis/IsoLight System for single-cell proteomics/phosphoproteomics, and several other analytical shared instruments for the analysis of proteins, lipids, and other biomolecules from different sources and organisms.
