Abstract
;为改善目前处于临床Ⅱ期中的乙肝病毒衣壳蛋白抑制剂甲磺酸莫非塞定(GLS4)水溶性差、口服生物利用度低的缺点,设计合成了一种前药,并且进行了前药的体外研究,包括细胞毒性和细胞水平抗HBV4效应(抑制HBV DNA复制和HBsAg、HBeAg分泌);进行了前药的水溶性、小鼠急性毒性及大鼠单剂量口服和静脉药代动力学研究.与GLS4相比,所设计的前药具有相似的抗HBV活性,对HepG2.2.15细胞的细胞毒性显著降低,水溶性增强,急性毒性较弱,但体内研究不稳定.
