Isolation Of Genomic Dna From Myrciaria Dubia (Hbk) " Camu Camu" Suitable For Molecular Analysis

Myrciaria dubia "camu camu", a native species of the Amazon that produces fruits high in vitamin C and other important substances. However, molecular studies of this plant are scarce, for lack of a reproducible protocol to purify their nucleic acids. The aim of this study was to establish a protocol to isolate genomic DNA from leaves of M. dubia, suitable for molecular analysis. DNA was purified with a modified protocol, the quality and quantity was estimated by spectrophotometry and agarose gel electrophoresis. Additionally, the quality was assessed by RAPD. The mean ratio of quality (A(260)/A(280)) of DNA was 1.9 +/- 0.1. By means of electrophoresis the DNA was intact and without RNA. Also, the synthesis of RAPD amplicons suggests absence of inhibitors for polymerases. The average concentration of DNA was 99 +/- 33 ng/mu l and the average yield was 237 +/- 80 mu g ADN/g leaf. In conclusion, was established a protocol for isolation genomic DNA from leaves of Myrciaria dubia "camu camu", characterized for allowing us obtain DNA of high quality and quantity sufficient for molecular analysis as RAPD.
