丹参酮ⅡA脂质体的制备及对HepG2/ADR细胞增殖的抑制作用

Abstract
目的 研制丹参酮ⅡA(Tan ⅡA)脂质体,考察其对人肝癌HepG2/ADR阿霉素耐药细胞增殖的抑制作用.方法 采用薄膜分散法制备Tan ⅡA脂质体,改良MTT法测定药物肿瘤细胞增殖的抑制作用.结果 制备载药脂质体的平均粒径为68nm,Zeta电位为-23.4 mV;载药量与包封率分别为138.9 ±8.7 μg·mL-1、97.4%±1.2%(n=3);与游离药物相比,Tan ⅡA脂质体对HepG2/ADR的增殖细胞有更强的抑制(P<0.05),并呈时间和剂量依赖性.结论 Tan ⅡA脂质体具有治疗多药耐药肝癌的前景.
