•Two chromatographic methods (UHPLC and UHPSFC) were developed and validated for determination of agomelatine and its impurities.•Selectivity of different additives in UHPSFC mobile phase was studied.•In UHPLC, better separation selectivity was obtained at basic pH.•Both methods were used for tablet samples analysis.•Detailed comparison of UHPLC and UHPSFC methods was made.
