•We provide new insights into the insulin response in cows with COD.•Lower gene and protein expressions were detected of some insulin downstream effectors.•Alterations detected would negatively influence the functionality of the ovaries.•Results obtained contribute to knowledge of the dynamics of the follicular persistence.
