This study presents the pioneering co-structure of HSV-1 gB and its neutralizing antibody D48, offering not only a structural foundation for D48 neutralizing HSV-1 infection but also revealing a potentially conserved antigenic domain gB DII, across the herpesvirus family, spanning pre- and post-fusion configurations. This lends support to the proposition that DII domain holds a pivotal and shared role during viral infection, presenting an auspicious target for the development of broad-spectrum drugs across the herpesvirus family.
