Cells were transfected with the indicated shRNA for 48 hr and immunostained with BAP1 antibody and counterstained with DAPI to reveal DNA. (B) Transfection of the FokI nuclease fusion construct into U2OS cells containing the stably integrated reporter leads to local accumulation of damage response proteins γH2AX and GFP-BAP1. 24 hours after co-transfection of GFP-BAP1 and Fok1 constructs, cells were immunostained with γH2AX antibody and counterstained with DAPI to reveal DNA.
